研究概要	1.スペルミジン取り込み系のペリプラズムに存在する基質(スペルミジン)結合蛋白質のスペルミジン結合部位を、X線結晶構造解析及び1アミノ酸残基置換変異PotD蛋白質の活性測定より同定した。その結果、4分子の酸性アミノ酸残基、3分子のTrp残基及び3分子のTyr残基がスペルミジン結合に関与していることが明らかとなった。なかでもNH基を認識するAsp257が最も重要であった。また、スペルミジン[NH_2(CH_2)_4-NH(CH_2)_3NH_2]のアミノプロピル基を認識するアミノ酸残基(Glu171及びTrp255)がPotDによるスペルミジン認識に強く関与していた。プトレスシン排出を行うPotEの構造と機能を解析した。PotEはプトレスシンが細胞内に過剰蓄積すると、プトレスシン/オルニチンアンチポ-タによりプトレスシンを排出した。プトレスシンに対するKm値は73μMであり、反応はエネルギーを必要としなかった。一方、PotEはプトレスシンを取り込むこともできた。この場合はエネルギー源として膜電位を必要とし、プトレスシンに対するKm値は1.8μMであった。PotEのトポロジーをPotEの長さの異なるN-末端部分にアルカリフォスファターゼ(AP)またはβ-ガラクトシダーゼ(β-gal)を融合し、活性を測定することにより決定した。APはペリプラズムに存在する場合のみ活性を示し、β-galは細胞質に存在する場合のみ活性を示す。この活性測定より、PotEは12ヶ所の膜貫通領域を持ち、N末端側とC末端側は共に細胞質側に位置することが明らかとなった。また、PotEの活性中心は細胞質側に存在し、Glu77、207、433が活性発現に必須であった。
