研究概要	組換え可溶性組織因子と活性型凝固第VIIa因子の複合体の結晶解析は、小さいながらも結晶が得られている。現在、X線結晶解析に使用する大きさの結晶の成長を待っている状況にある。Max-Planck研究所でその解析を進めている。組織因子の細胞外領域を構成する2つのドメインの役割に関して、幾つかの進展をみた。2つのドメイン(NドメインとCドメインとよぶ)を単離するため、両ドメイン間に凝固Xa因子の認識配列を挿入し、酵母で発現させた。精製後、本認識部位で切断し、NドメインとCドメインをカラムクロマトグラフィーで単離した。各ドメインによる凝固VIIa因子のアミダーゼ活性の増幅能を調べたところ、Nドメインには全く増幅能が認められなかったものの、Cドメインは約5%の活性が認められた。一方、両ドメインを混合後、活性を測定すると、切断前の活性にまで回復したことから、組織因子による凝固VIIa因子の合成基質に対する活性の増幅には、組織因子を構成する2つのドメインが必要であり、NおよびCドメイン単独ではその活性が極めて弱いことが判明した。我々は既にヒト血漿中の活性型凝固VIIa因子の定量法を確立している。血漿中には総VII因子の0.5-1%に相当する2-4ng/mlの活性型VIIa因子が循環している。生体内に於けるVIIa因子の活性化機序を明らかにするため、各種病態下のVIIa因子量を測定したところ、冠動脈疾患や慢性脳梗塞で増加がみられた。糖尿病患者でも増加がみられた。高血圧患者では、微量アルブミン尿を伴う場合はVIIa因子量の増加がみられたが、伴わない場合はVIIa因子量は増加していなっかた。尿中アルブミン排泄量の増加とともに、VIIa因子量が増加することを考えると、腎障害とVIIa因子の生成は極めて密接な関連があると推測された。
