癌抑制遺伝子RBを中心とした細胞周期の解析

研究概要	癌抑制遺伝子RBは、細胞の増殖停止を引き起こすので細胞中での過剰発現が困難であったが、RBを誘導発現させると増殖を停止するNIH3T3由来細胞系を作製した。また、血清ロット依存性が判明し、過剰発現で増殖を完全に停止させる血清とほとんど増殖に影響しない血清を捜し当てることができた。これは、条件感受性突然変異に類似しておりこの系の有用性を高めている。また、生命の基本設計図はすべて、遺伝子として記録保持されており、転写というこの遺伝子情報を取り出して具現化する作業は生命現象の根幹をなしている。Differential display法(DD法)は転写産物を効率、再現性良く記述する方法であり、生命現象の変化にともなう転写産物の発現量の変化を解析するには最適の方法である。RB産物は、転写因子などと相互作用することによって、細胞の転写状態の調節を通して機能を発揮していることが示唆されているので、RB強制発現の結果は、細胞内の転写産物の量的変化に反映され、その解析には、DD法が最適である。また、DD法はその遺伝子の生物学的意義、実用性の有無等の判別が短期間で可能で、無駄な実験を省きやすい。DD法に種々の改良を施し、再現性と効率をさらに高めた独自の改良DD法を、本研究のRB細胞に適用し、RBの発現によって転写量の変化する遺伝子のcDNAの単離、解析、同定をなお押し進めている。また、現在、細胞の接触阻害、血清飢餓、血清刺激、senescence、神経細胞の分化、アポトーシスに重要な遺伝子を同法で単離、同定、解析している。これらの現象を手広く解析することは、同じ増殖停止にともなう転写量変化の中でも、RB発現、分化、血清飢餓、接触阻害、老化などに共通なもの、特異的なものの分類に非常に有用である。これらを総合した結果を得るには更なる研究と期間が必要である。
