ミトコンドリアにおけるヘム合成終末酵素群の複合体形成および相互作用に関する研究

研究概要	動物のヘム合成系の最後の三段階はコプロポルフィリノーゲンオキシダーゼ(CPOX)、プロトポルフィリノーゲンオキシダーゼ(PPOX)およびフェロケラターゼ(FECH)と呼ばれている終末酵素群として知られており、それぞれミトコンドリアに局在している。本研究では、これらの三酵素のミトコンドリア内における詳細な分布、酵素間のポルフィリン輸送調節機構を明らかにするために酵素群複合体の形成の有無、ポルフィリン輸送と鉄代謝間の調節および赤血球分化時のヘム合成の誘導に伴う3酵素の発現機構ならびに律速段階を支配する因子等について、クローン化したCPOX,PPOXおよびFECHを用いて調べた。本研究は赤血球分化時のCPOXおよびFECHの誘導発現について、それぞれの遺伝子の5-プロモーター領域を用いたプロモーターアッセイを行った。その結果、CPOXの発現および誘導にはSp-1とGATA-1の関与が泌須であることが解ったが、FECHのそれらには、GATA-1ではなく、むしろTCGCCC配列を有するSp-1様superfamilyが関与することが解かり、ふたつの酵素の発現は異なることを明らかにした。ミトコンドリアにおけるポルフィリン輸送に関与すると考えられる末梢型ベンゾジアゼピン受容体は、ヘムに対する親和性をも有しており、合成されたヘムのミトコンドリアからの搬出にも関与していることが解った。また、神経細胞に特異的に発現する本受容体のホモログp24を単離した。p24蛋白質の機能や性質の詳細については未だ不明な点が多く、現在ヘム合成との関係も含めて検討している。また、20kDaのFECH会合蛋白質のcDNAを単離した。20kDa蛋白質はひとつの膜貫通部位を有し、赤血球分化時に誘導されるがその他の性質については不明である。今後さらに、その機能について検討する。
