ホヤ卵細胞質に存在する表皮及び内胚葉決定因子の精製

研究概要

ホヤの幼生においてアルカリ性フォスファターゼ(AP)活性が内胚葉細胞において検出されることが示されている。過去の報告では、転写阻害剤であるactinomycinDを受精後処理し続けた場合でも幼生期にはAP活性が認められることから、AP活性の発現には、zygoticな転写は必要なく、卵内に存在するmaternalなAP mRNAの翻訳によりなされることが示唆されていた。今回、maternalなAP mRNAが卵内に存在しているかどうかを、直接的に確かめるため、マボヤ(Halocynthia roretzi)から内胚葉特異的なAPを精製し、そのN末端アミノ酸配列を決定し、更にこの配列をもとにcDNAを単離し、全配列を決定した。単離したcDNAは完全なORFとポリA付加シグナルをもつ3017bpのクローンで、マウスのtissue-nonspecific型APと52.6%のホモロジーを持ち、マボヤの尾芽胚期において内胚葉特異的に発現することが確かめられた。Northern blotとin situ hybridizationの結果、APのmRNAは未受精卵には検出されず、神経胚期においてはじめて内胚葉前駆細胞に検出が認められた。これらの結果から、少なくともマボヤにおいては、内胚葉特異的なAP活性の発現には卵内に存在していると考えられる細胞質決定因子によるzygoticな遺伝子発現が必要であることが示された。
