研究概要

我々は、まずCalmegin cDNAの一部をプローブとして129ゲノムライブラリーより転写開始点から約3kb上流の遺伝子を調整した。この遺伝子の様々な欠損ミュータントにレポータージーンとしてCAT遺伝子をつなぎ、Calmegin遺伝子を発現しているマウス精巣性テラトカルシノーマF9細胞にトランスフェクションして転写活性を調べた結果、転写開始点より上流200bpという短い領域にその特異的な転写活性を認めた。また、これらのレポータープラスミドを導入したトランスジェニックマウスを作出し解析したところ、同領域がin vivoの精巣での発現に必要な領域であることが認められた。
また精細胞特異抗体を用いて発現ライブラリーより抗原遺伝子を単離したところ、11種類の抗原遺伝子が単離され、ホモロジー検索により6種類が新規の遺伝子であり、既知の遺伝子と相同性をもつものが5個認められた。これらのうちGAG12は、癌抑制様遺伝子(gene 311)と相同性があり、また発現が減数分裂時に見られることから、生殖細胞の細胞周期制御においての機能に興味を抱かせるものであった。さらに我々のクローニングした減数分裂特異的遺伝子についてヒト精巣RNAのノーザンブロッティングを行ったところ、ヒト相同遺伝子の存在が示唆された。これらのうちSCP遺伝子についてはヒト精巣cDNAライブラリーよりhSCP遺伝子の単離を行い、塩基配列を決定した。
さらに、p53欠損マウスより精細胞の株化を行ったところ、何株か不死化した細胞株が得られた。そこで、各種モノクローナル抗体を用いて分類したところセルトリ細胞株であり、精細胞株は得られなかった。
