葉形態形成機構の発生遺伝学的解析

研究概要	陸上植物のシュートの基本パ-ツである葉は、植物の形態形成の機構を知る上で最も重要な要素である。しかしその発生過程の詳細はまだ明らかになっていない。その原因は、発生に可塑生が大きく、また葉の発生そのものが現象論的に複雑なところにある。双子葉植物の葉の発生は、性格の相異なる事象を同時かつ同所的に伴うため、解剖学的には解析が困難であった。これを打開するためには材料としてアラビドプシス(Arbidopsis thaliana (L.) Heynh.)の変異体を用い、この複雑な葉の発生過程を現象論と切り離し、遺伝学的な素過程に分割するのが有効と考えられる(Tsukaya et al. 1993, Tsukaya et al. 1994, Tsukaya, 1995a, 1995b)。今年度はこれまでの解析に続き、今まで単離・収集してきた葉の形態に関する突然変異体を材料に、解剖学的・遺伝学的解析を行った。その結果、葉の細胞の極性伸長を司るANGUSTIFOLIA, ROTUNDIFOLIA3各遺伝子(Tsuge et al., in press)、極性によらず細胞伸長を司るACAULIS1遺伝子、細胞の伸長と分裂の両方を司るLANCEOLATUS1遺伝子、さらに細胞の極性伸長を抑制するMACROPHYLLA遺伝子が同定できた(Tsukaya et al., 1995)。また葉の形状が非対称的になる表現型を示す変異体として、as1, as2を含むdeformifolia変異体を同定した。これらは互いに独立に機能しているものと推察される。またT-DNAの挿入による変異株を探索し、4種の候補を得た。さらに、変異体の細胞学的解析のための新たな技術として、アラビドプシスの葉の光学的な組織解剖法を見出した。来年度は、以上の成果を元に、葉の分子/細胞レベルでの総合的な形態構築解析をさらにすすめ、植物の葉を構築する未知の制御系を解明したいと考えている。
