研究概要

核多角体病ウイルス感染初期反応時のウイルスと細胞との相互作用を理解することを目的として、ウイルスレセプターの同定、ならびにタンパク質リン酸化の関与について検討した。
1.ウイルスレセプターの同定
宿主細胞を抗原としてマウスを免疫し、細胞表面抗原を認識する抗体を作製した。免疫マウスの脾臓とミエローマ細胞を融合してハイブリドーマを作製し、ウイルス感染に対して中和活性能力を持つハイブリドーマをクローン化するための、中和活性能力の検定法を検討している。
2.ウイルス感染過程におけるタンパク質リン酸化の関与について
7種類のタンパク質リン酸化阻害剤がウイルス感染に及ぼす影響を、(1)プラークアッセイ法による合成ウイルス量の検定、(2)多角体形成様相の顕微観察、(3)ウエスタンブロット法による合成ウイルスタンパク質の検出により調査した。その結果、Cキナーゼの阻害剤であるH-7によって、ウイルス感染の初期過程が遅延することが明らかとなった。また、宿主細胞の増殖をMTT法により調査した結果、H-7によってウイルス感染による細胞増殖の停止が抑制された。すなわち、細胞はウイルス感染によって増殖を停止するが、H-7処理することによって、ウイルス添加後も増殖する傾向を示した。現在、ウイルス感染初期過程のどの段階でリン酸化反応が関与しているのかを検討中である。
