研究概要	当初の研究実施計画にしたがい、まず約20種のSSI様インヒビター(SIL)生産菌の培養上清より精製単離したSILタンパク質の全一次構造決定と阻害特性の解析を行った。その結果、SSIの機能構造を形成する上で重要と考えられた部分は高度に保存されていることがわかった。また、標的プロテアーゼの基質特異性と阻害反応部位近傍の配列との間に強い相関性のあることがわかった。さらに、インヒビターの構造相同性と既知の生産菌種間の近縁関係と良い対応があったので、SILインヒビターの構造相同性を利用し、最大節約法と近隣結合法を用いて進化系統樹を作成した。次に、SSI消失変異株を取得したところ、胞子形成能の低下、生育速度の低下、プロテアーゼ生産の増大など多面的形質の変化が観察された。そこで、SSIと相互作用する菌体外の内因性標的プロテアーゼをアフィニティーカラムにて複数単離した。その一つ、SAM-P20の遺伝子クローニング、塩基配列決定を行った。その結果、推定アミノ酸配列が既知のS.griseus proteaseAおよびBのそれらと高い相同性のあることがわかった。また、組換え大量生産により得た完全精製SAM-P20の酵素化学的解析を詳細に行った。特筆すべきは、基質特異性がキモトリプシン様である上に、トリプシン様の性質も合わせ持つ点である。また、SAM-P20のプロテアーゼ活性は明らかにSSIによって阻害を受けることがインピトロ実験において明らかとなっった。今後は、SAM-P20とSSIとの複合体形成の詳細な解析を実施する予定である。他の内因性標的酵素についても同様の研究を推進していくことを考えている。
