プラスミド接合伝達活性化ペプチド"細菌の性フェロモン"の受容認識機構

研究概要	本研究では、腸球菌Enterococcus faecalisの性フェロモン応答性接合伝達プラスミドにコードされる、フェロモンの認識およびシグナル伝達機構を分子構造レベルで解明することを最終目標とし研究し、以下の二研究を行った。 (1)性フェロモンのシグナル伝達の遺伝的解析性フェロモンのシグナル伝達に関与していると思われる領域には、複数のタンパク質(tra遺伝子産物)がコードされている。本研究では、相同組換えによる変異導入法を用い、個々のtra遺伝子に変異を導入し、個々の遺伝子が性フェロモンのシグナル伝達にどのような役割を果たしているかを解析した。その結果、フェロモンの生産を制御するフェロモンシャットダウン遺伝子traB、フェロモンに対する感受性を担うtraC、およびフェロモン応答のネガティブレギュレーターtraA遺伝子を同定できた。それらの変異を掛け合わせることにより、それらの性フェロモンシグナリングにおける上流、下流の相対的位置付けを行なった結果、traB-traC-traAの順に位置付けされた。 (2)フェロモンの追跡実験追跡実験用にまずフェロモンのラジオアイソトープラベルを試みた。フェロモンの一種cAM373のN末端アラニンをチロシンに変換しそのチロシン残基に^<125>Iを導入したフェロモンアナログは、供与菌に投与すると短時間に分解され、このアナログは、フェロモンの追跡実験には不適当であることが判明した。そこで次に他種フェロモンcPD1のロイシン残基に接触還元を用いてトリチウムを導入した。この^3H-cPD1は供与菌細胞表層に特異的に結合した。現在、このラベルフェロモンを用いた追跡実験をさらに進めている。
