下垂体後葉における神経分泌果粒の輸送と放出の分子機構

研究概要

1.私が急速凍結エッチングレプリカ法によって発見した、下垂体後葉軸索内の微細管と分泌果粒間の架橋構造は、神経分泌果粒の輸送に重要な役割を果たしていると考えられる。一般の神経の軸索においては、順行性に運ばれる膜小器官と微細管の間の架橋構造はモータータンパクのkinesinであることがわかっている。本研究において、免疫組織化学法によって下垂体後葉にkinesinがかなり大量に存在することがわかり、さらに免疫電顕法によって軸索内の神経分泌果粒にkinesinがassociateしていることが確認され、神経分泌果粒と微細管の間の架橋構造がkinesinである可能性が示された。
急速凍結エッチングレプリカ法で観察すると、下垂体後葉の軸索内を走る微細管束はたくさんの架橋線維で束ねられている。本研究によって、これらの架橋線維がMAPa(特にMAP1とタウ)であることが確認された。
3.ウェルシュ菌のエンテロトキシンは神経筋伝達をブロックする。本研究によって、エンテロトキシンが神経筋接合部のpresynaptic nerve terminalに結合し、synaptic vesiclesを枯渇させることなく神経筋伝達をブロックすることがわかった。このことから、エンテロトキシンがsynaptic vesicleのエキソサイトーシスをブロックしていることが推測される。ウエルシュ菌のエンテロトキシンはエキソサイトーシスの機構を解明するための有用なツールとなりうるであろう。
