研究概要

中枢神経系における内向き整流性カリウムチャネルの分子機構を明らかにするために、マウス脳よりまったくあたらしい2回膜貫通型内向き整流性カリウムチャネルcDNA(KAB-2,the second type of inward rectifying K^+ channel with an ATP-binding domain)をクローニングした。アフリカツメガエル卵母細胞発現系でKAB-2チャネルは、古典的な内向き整流特性を示すカリウム電流を示した。in situ hybridization法による解析の結果、KAB-2mRNAは脳幹部神経核とともに、脳梁や小脳白質などのグリア細胞に強く発現していた。免疫組織化学の結果、アストロサイト初代培養にKAB-2チャネルが発現した。また、KAB-2チャネルは胎児期脳には発現が少なく生後1週でその発現が著しく増加し、生後2週で成体レベルに達する。この発現パターンは生後脳内の神経ネットワークの形成とともにグリア細胞の増殖していくことと一致する。以上のことから、KAB-2チャネルはグリア細胞におけるK^+-buffering systemに関与していると強く示唆される。現在、生体内、特にグリア細胞K^+-buffering systemにおけるKAB-2チャネルの生理的役割を明らかにするために、ES細胞を用いた発生工学的手法によりKAB-2チャネルのノックアウトマウスの作成を試みている。
