低分子量G蛋白質Rhoの活性化機構と作用機構

研究概要

本研究では、Rho低分子量G蛋白質の活性化機構、機能および作用機構について検討した。私共を含めたいくつかのグループによって、Rhoが、アクチン細胞骨格の制御を介して、種々の細胞機能を制御していることを見い出している。本年度の研究において、まず、RhoをADPリボシル化して不活性化するC3を用いて、Rhoの新しい細胞機能についてさらに検討した。その結果、MDCK細胞において、細胞外Ca^<2+>濃度の上昇によって細胞間接着(adherens junctionおよびtight junctionの形成)が誘起されるが、RhoはそのマーカーであるE-カドヘリンのadherens junctionへの局在、および、ZO-1のtight junctionへの局在に関与していることが明らかとなった。一方、細胞膜上でアクチン細胞骨格の再構成を制御する蛋白質系としてERMファミリー(Ezrin,Radixin,Moesin)-CD44系が知られているが、私共は、すでに、Rhoが細胞内でERMと共に局在していることを明らかにしている。しかし、RhoがERMの機能を制御しているか否かについては不明であった。そこで、本年度の研究において、C3をマイクロインジェクションしたMDCK細胞において、ERMの局在を検討したところ、RhoはERMの細胞辺縁領域への局在を制御していることが明らかとなり、RhoがERMを介してアクチン細胞骨格を制御していることが強く示唆された。今後は、Rhoのこの機能に関与する標的蛋白質を単離し、それがどのような機構でERM-CD44系を介してアクチン細胞骨格の再編成を制御しているのかを明らかにする予定である。以上の結果を得たことにより、本年度の本研究の研究計画はほぼ達成できたと考えている。
