エンドセリン-1の眼圧血液房水柵に与える影響とその眼内動態及び受容体

研究概要

1.エンドセリン(ET)-1 0.5μgを1眼に他眼にプラセボを硝子体内投与後 眼圧を経時的に測定した所、0.5〜2hrで一過性眼圧上昇をした後、72hr以上つづく 持続性眼圧下降を認めた。また、ET-1投与1,24,48hr後の房水中のPGE_2,蛋白濃度とも有意に上昇していた。
2.ET受容体のカブタイプであるET_A受容体の選択的拮抗剤97-139 155μgを硝子体内投与後ET-1(0.5μg)を両眼に投与した所 ET-1による眼圧上昇 眼圧下降ともに有意に抑制された。また、房水中のPGE_2、蛋白濃度の上昇も有意に抑制された。
3.ET受容体のサブタイプであるET_B受容体の選択的刺激剤snrafotoxin S6_C0.5μgを硝子体内投与後 眼圧は一過性上昇を認めず、96hrに及ぶ有意の下降のみ認めた。また、房水中のPGE_2、蛋白濃度は有意に上昇した。
4.ET-1(0.5μg)硝子体内投与後、眼球各部位(毛様体、虹彩、硝子体、前房、網脈絡膜)の組織内濃度測定した所、前房、毛様体、虹彩へのET-1の移行は網膜に比して軽微であることが判明した。
I^<125>-ET-1(0.5μg)を硝子体内投与後 毛様体上皮、前房隅角線維格帯受容体に結合しつづけていることが判明した。
