コレラトキシン結合SIV構成蛋白の経口免疫による腸管粘膜内細胞障害性T細胞の誘導

研究概要	我々の研究室ではヒトAIDSの動物モデルであるrhesus monkeyにSIVmacを感染させると、AIDSウィルス構成蛋白gagおよびenvに対するcytotoxicT細胞(CTL)活性が早期に高くなるものでは発症が遅く、逆に活性が低いものでは早期に発症することを報告してきた。今回の研究はウィルス構成蛋白の経口免疫によるAIDSウィルス特異的CTLを誘導する新しい治療法の開発を目的とする。本年度は、樹立したSIVmac感染rhesus monkey腸管粘膜内CTL活性に及ぼすサイトカインの役割について検討した。CTLの機能発現の最終段階にIL-2が必要であると信じられている。一方、種々ウイルス感染症においてCTLがウイルス排除に重要な役割を担っていると考えられている。以上の観点から、すでに、米国ではNIHが中心に、エイズ患者に対し、ヒト・ricombinant・IL-2の全身投与が行われた。しかし、この臨床治験においてCTL活性の上昇は認められたが、逆に、ウイルスのreplicationも上昇してしまい、臨床治験はストップした。今回、我々は最近、発見されたIL-15がIL-2と共通のレセプターを介して作用するが、その産生細胞はまったく異なる。特に、腸管上皮細胞はIL-15を産生するが、IL-2を産生しないことに着目し、IL-15による腸管粘膜内CTLクローンの活性増強作用を証明した。一方、SIVgagに対するDNAワクチンにより、rhesus monkeyにCD8陽性CTLが誘導されることを証明した。DNAワクチンは筋注とされ、筋細胞はIL-15を代表的産生細胞であり、このことからもIL-15による腸管粘膜内CTLクローンの活性増強作用は十分期待できる。今後、経口ワクチンによるHIVウイルス防御戦略において重要なステップと考える。
