バイオテクノロジーを利用した好酸球顆粒蛋白質の大量調整と好酸球機能及び分化の解析

研究概要

[1]好酸球増多症モデルを活用してラットmajor basic protein (MBP)の精製行い、部分アミノ酸配列を明らかにした。さらに、得られた情報を基に、MBPのcDNAクローニングを以下に述べる方法で行った。好酸球増多症を誘発させたラットの骨髄から骨髄細胞を採取し、AGPC法によりtotal RNAを調整し、RT-PCR法によってMBPのcDNAの部分塩基配列を決定した。その結果、約240bpのPCR産物が得られ、その塩基配列にはヒトおよびモルモットのMBPの対応する部分と高い相同性が認められた。これらの塩基配列をもとにプライマーを作製し、3'および5'RACE法によりラットMBP cDNAの全長の塩基配列を決定した。ラットMBPのcDNAは全長が867bpで5'noncoding regionが45bp、coding regionが684bp (signal peptide、proprotein、mature MBPの合計領域)、3'coding regionが125bp (poly A tailを含む)であった。現在、RT-PCR法によりラットMBPの全長のcDNAをもつプラスミドを作製し、recombinant MBPを作製するための準備を行っている。
[2]精製したラットMBPをウサギに投与することにより、極めて特異性の高い抗体を得ることに成功した。本抗体を用いることにより、ラットを抗原で感作すると、骨髄中にMBP抗体に結合する蛋白質が増加することが明らかになった。また、[1]で作成したプライマーを用いてRT-PCRを行った結果、骨髄細胞中のMBP mRNA量は、感作をすることにより上昇することが明らかになった。従って、ラットを抗原で感作すると、骨髄中で好酸球の分化が亢進し、好酸球増多の準備段階が進行することが示唆された。
