研究概要	Bromodomainはグローバルに作用する新しいタイプの転写因子に広く含まれている構造ドメインで転写因子間相互作用に関わり、クロマチン転写などに働くものであると考えられている。一方、TFIIDはクロマチン転写に関わると考えられているもののどのような段階にどのように関わるかは明らかでないが、ここに示したBromodomainをもつサブユニットCCG1を持つ。したがって、このBromodomainを手がかりにTFIIDの持つクロマチン転写における機能的役割を明らかにする目的で以下の解析を進めた。 1)Bromodomainと相互作用する新しいタイプの因子をコードするcDNAの単離 Bromodomainと相互作用する因子をコードするcDNAを蛋白質-蛋白質相互作用活性を指標にしたスクリーニング法、すなわちtwo-hybrid法により単離した。そのうち、5種は未知因子であったが2種の因子は互いに似ているばかりでなく、出芽酵母より遺伝学的に単離されていた因子のヒトホモログでクロマチン転写に関わるであろう性質を持っていた。 2)Bromodomainと相互作用する活性で単離した因子の性状解析単離した上述の2種の因子とCCG1のBromodomainとの相互作用の特異性検定後、この因子の局在性や発現パターンを調べたところ核内に存在すること、一方はどの組織でも発現しているが、他方は幾つかの組織でしか発現しておらず、組織特異的な活性を担い得る因子であろうことが判明した。しかもこの因子は既存の転写因子の構造モティーフを持たないことから、新しいタイプの転写調節反応に関わるであろうことが示唆された。
