新規なキノン系補酵素類の機能解明とその応用

研究概要	本研究では最近になって次々と発見された全く新しいキノン系の補酵素に着目し、その機能解明と応用を目的とし主にモデル化学的な立場から検討を行った。研究対象とした補酵素は、新規な複素多環オルトキノン系の構造を有するPQQ(Pyrroloquinolinequinone)、TTQ(Tryptophan Tryptophylquinone)、およびチロシンから誘導されたと考えられているTyr-Cysである。【1】補酵素PQQの酸化還元過程の解明:申請者らはこれまでに補酵素PQQの基本的な物性や化学的性質について詳細に検討を行なうとともに、種々の生体関連基質、特にアミン、アミノ酸、ヒドラジン、糖類、チオール、NADHなどの効率的な触媒的酸化反応を見い出してきた。本研究では、それらの詳細な反応機構や還元型PQQの再酸化過程、あるいは金属イオンとの相互作用や光化学的挙動などを明らかにするため、申請者らがこれまでに合成してきた数十種類におよぶPQQのモデル化合物あるいは誘導体の化学的説質や機能について、系統的に検討を行った。【2】新規補欠分子TTQの化学的機能の解明:新しい補酵素TTQの基本的な物性やアミンの酸化反応機構および構造活性相関を解明するため、種々のTTQモデルを合成し、それらの物性やアミン酸化機能について系統的に検討を行うとともに、生体系におけるTTQの役割について考察を加えた。【3】ガラクトース酸化酵素活性中心のモデル化:ガラクトース酸化酵素の活性中心から見いだされた全く新しいタイプの有機補欠分子であるTyr-Cysのモデル化合物を種々合成し、それらの物性や酸化還元挙動、および銅イオンとの相互作用などについて詳細に検討し、酵素活性中心のモデル構築に成功した。
