研究概要

本研究の目的は、神経系、免疫系などの高次生体システムの機能発現と機能制御の機構、ならびに発生過程における高次生体システムの構築機構を明らかにし、高次生体システムの基本原理と相互に関連した制御機構を明らかにすることである。
平成7年度の研究は、以下に述べるように高次生体システムの機能制御の機構に関して多くの成果を挙げ、平成8年度以降の研究の重要な礎を作った。
1.中西のグループは、種々のグルタミン酸受容体ノックアウトマウスの作製に成功し、(1)網膜のmGluR6の欠損は明暗の識別、瞳光反応の低下をもたらすこと、(2)mGluR2の欠損は海馬のLTDの誘導がなくなるが、場所の記憶は阻害されないこと、即ち海馬の可塑性と記憶には解離がみられること、(3)NMDA受容体のNR2A,NR2Bサブユニット単独の欠損は運動障害をおこさないが、両者が同時に欠損する事によって小脳由来の運動障害がおこること等、記憶、学習、外部刺激の情報処理の機構に関し、多くの事実を明らかにした。(4)NMDA受容体およびmGluRの細胞内Ca^<2+>伝達機構及びグルタミン酸による細胞死の機構に関しても新事実を明らかにした。
2.本庶のグループは、(1)ES細胞とstroma細胞との共培養により血球系へ分化する系を活用して血球系分化の初期段階における増殖因子及び膜発現レセプターの多数をsignal sequence trap (SST) 法を用いて単離することに成功した。(2)細胞死に際して発現される新しい遺伝子PD1が、胸腺内における特定のTリンパ球分化段階に極めて短時間特異的に発現することを明らかにし、PD1のノックアウトマウスの完成とあわせて、今後この分子の生理的な意義を明らかにする重要な礎を作った。(3)リンパ節形成不全マウスにおいては、胚中心が形成されず、クラススイッチやsomatic mutationが著しく阻害されていることを明らかにし、免疫応答における場の重要性を明確に証明した。(4)神経系や免疫系の分化制御に関わるRBP-J_Kが膜受容体notchと新しいdomain RAM23を介して、直接結合することを証明し、notchレセプターの各一部が切断され、核での転写制御を行うという新たな機構を示唆する結果を得た。
3.西川のグループは、(1)多分化能を有する未熟胚細胞やES細胞から、血液・血管細胞や神経芽細胞への分化を誘導する培養条件を確立した。(2)原条上皮細胞から分化してくる中胚葉を4種類に分類できる細胞表面マーカーを明らかにした。(3)これを用いて、初期胚やES細胞の培養から将来の運命別に中胚葉細胞を分取する技術を確立した。(4)新たにFlk2,PDGFRβの作用を阻害するブロック抗体の作製に成功し、抗PDGFRβ抗体投与により、血管形成を中心に多様な発生異常が誘導された事を明らかにし、又、抗PDGFRα抗体投与の実験から、網膜でのgliovascularネットワークの存在を証明した。
4.西田のグループは、(1)MAPキナーゼカスケードが、Xenopus初期胚の中胚葉誘導過程に必須の役割を果たしている事を示し、又この時のMAPキナーゼカスケードの作用点が、FGFの下流であることを示した。(2)MAPキナーゼスーパーファミリーの一つであるp38の活性化因子として、MAPKK6を新規にクローニングした。(3)MAPKK-Kの新規メンバーとしてTAK1を同定し、TGFβとBMPの刺激により活性化することを示した。(4)p38の別個の活性化因子であるMKK3とMAPKK6はいずれも、TAK1により直接にリン酸化された活性化されることを明らかにし、TAK1-MKK3/MAPKK6-p38という新しいキナーゼカスケードの存在を示唆した。
