研究概要	本研究の目的は、神経系、免疫系などの高次生体システムの機能発現と機能制御の機構、ならびに発生過程における高次生体システムの構築機構を明らかにし、高次生体システムの基本原理と相互に関連した制御機構を明らかにすることである。平成8年度の研究は、以下に述べるようにCOE拠点化の研究体制のもとに高次生体システムの機能制御の機構に関して多くの成果を挙げ、そのいずれもが平成9年度以降の研究の発展につながるものである。 1.中西のグループは、種々のグルタミン酸受容体のノックアウトマウスの作製に成功し、(1)メタボトロピック受容体(mGluR)の一つmGluR6が明るさに反応する受容体として働き、mGluRとAMPA受容体が巧妙に使い分けられることによって明暗が識別されていること、(2)他のmGluRサブタイプ(mGluR2)が海馬の神経可塑性に必須の役割を果たしていること、(3)小脳顆粒細胞のNMDA受容体のNR2A、NR2Cサブユニットが筋肉運動の調整に重要であること、(4)プロテインキナーゼCによる燐酸化がmGluRのCa^<2+>oscillationを決定していることを明らかにした。 2.本庶のグループは、(1)Tリンパ球の初期分化の一時期と抗原刺激後の一時期に特異的に発現するPD1遺伝子のノックアウトマウスの解析を行い、この分子が胸腺内における新たなステップのTリンパ球選択に関与する可能性を明らかにした。(2)抗原刺激後のクラススイッチ遺伝子組換えに関わる遺伝子を明らかにするための新しいサブトラクション法を開発し、この方法により新たな数種類の遺伝子のクローン化に成功した。(3)Notch/RBP-Jレセプターシグナル伝達系において、その制御に関わる新たな分子Kyo-T2を発見し、これがNotchシグナルの抑制に関わることを明らかにし、さらにRBP-Jと類似の構造を持つ新たなメンバーとしてのRBP-Lの存在を明らかにした。 3.西川のグループは、(1)PDGFRファミリー分子の形態形成過程における機能について解析し、PDGFRαが成熟型造血支持組織の発生、胎盤の母体からの物質輸送に関わるDuval Sinusの発生、さらに神経系のニューロンの移動に必須であることを証明した。(2)運動別に中胚葉細胞を生きたまま純化する方法を開発した。(3)ES細胞から、近傍中胚葉を高率に誘導する培養方法を開発すると共に、その細胞をさらに血管・血液細胞へと分化させることに成功した。(4)血液幹細胞の増殖を、分化を停滞させたまま維持する無血清培養法を開発した。 4.西田のグループは、(1)Tリンパ球においてCD3及びCD28の共刺激によりMAPキナーゼスーパーファミリーの一つであるp38が活性化し、IL-2産生に関与することを示した。(2)小脳顆粒細胞においてグルタミン酸刺激によりp38が活性化し、アポトーシスに関与することを示した。(3)新規MAPKK-K、ASK-1を同定し、p38並びにSAPK/JNKの上流に位置することを明らかにし、さらにTNF-αによるアポトーシスに必要であることを示した。(4)MAPKKの細胞質局在が、核外移行シグナル配列(NES配列)によって規定されていることを見出した。
