研究概要

本研究の目的は、神経系、免疫系などの高次生体システムの機能発現と機能制御の機構、ならびに発生過程における高次生体システムの構築機構を明らかにし、高次生体システムの基本原理と相互に関連した制御機構を明らかにすることである。
平成9年度の研究は、以下に述べるようにCOE拠点化の研究体制のもとに高次生体システムの機能制御の機構に関して多くの成果を挙げ、そのいずれもが平成10年度以降の研究の発展につながるものである。
1.中西のグループは(1)トランスジェニックマウスの手法を用い、メタボトロピック・グルタミン酸受容体(mGluR)サブタイプ6が視覚の錐体と桿体の両系の光刺激を伝達する受容体であることを明らかにした。(2)mGluR6は明暗の識別に必須の受容体であることをmGluR6欠損マウスの動物行動学的解析によって明らかにした。(3)mGluR5がアストロサイトのCa^<2+>oscillationを決定していることを明らかにした。(4)mGluR2及びmGluR6の遺伝子発現調節機構を明らかにした。
2.本庶のグループは(1)抗赤血球抗体トランスジェニックマウスにおける自己免疫病の派生が腸管内細菌感染によるIL-5およびIL-10の分泌に依存することを明らかにした。(2)ヒトの抗体H鎖V領域遺伝子座の全領域(960キロベース)の塩基配列の決定と解析を完了した。(3)クラススイッチ制御の分子機構に関してミニ染色体を用いた解析を行い、クラス特異性を決めるのはS領域配列自身ではないことを証明した。(4)幹細胞の分化制御を行うNotch/RBP-Jシグナル伝達系の分子機構を追及し、Notch細胞内領域にRBP-Jと結合する2つのdomainとtransactivation domainとが存在することを証明した。
3.西川のグループは(1)パイエル板誘導に関わる細胞を単離し、同時に初期誘導過程の分子・細胞メカニズムを明らかにした。(2)中胚葉から血管、血液への分化過程で経過する中間段階を定義し、血液が血管内皮より分化することを初めて証明した。(3)c-Kitリガンドを皮膚に発現させたトランスジェニックマウスを作成し、色素細胞発生過程における膜型、分泌型リガンドの機能を明らかにした。(4)分化制御因子Id2が末梢リンパ組織、脳、乳腺の発生に必須であることを明らかにした。
4.西田のグループは(1)グルタミン酸刺激による小脳顆粒細胞のアポトーシスにMAPキナーゼ(MKK)スーパーファミリーの1つp38が関与することを明らかにした。(2)SAPK/JNKの活性化因子として新たにMKK7を同定し、TNPαなどの炎症性サイトカインによる遺伝子発現に関与することを明らかにした。(3)Fasのシグナル伝達の一部を、MKK7→SAPK/JNKおよびMKK6→p38の二つのキナーゼカスケードが担うことを明らかにした。(4)核外移行シグナル(NES)配列の受容体を同定し、CRM1であることを明らかにした。(5)MAPKKのNES配列が、MAPキナーゼの細胞内局在を規定しうること、細胞癌化を抑制することを明らかにした。
