研究概要

本研究は、脳における情報処理や神経伝達機構などの、生物の持つ優れた機能を持ち、且つ工学的に応用可能な全く新しい「生体機能分子」の「設計・合成」を行うことを目的とする。
生体機能分子を設計・合成するために、有機合成化学的アプローチ蛋白質化学的アプローチの2つの方針をとる。有機合成的アプローチとしては、モレキュラーインプリンティング法による分子認識部位の構築と触媒能の導入を行う。タンパク質工学的アプローチとしては、タンパク質の化学修飾による電子伝達効率の促進と、天然メディエーター分子の化学修飾による酸化還元特性の改変を行う。
平成7年度は具体的には以下の2つを目的として研究を遂行した。
1.原子、分子の立体配置をオングストロームレベルで精密に制御する技術としてモレキュラーインプリンティング法を確立し、分子識別機能のみならず、電子・電気機能や触媒機能をも同時に賦与した生体機能分子を創製する。
2.天然ポリペプチドや蛋白質と合成高分子あるいは無機材料をハイブリッド化し、電子伝達機能を持つ人工メティエーターを創製する。
1.については、モレキュラーインプリンティング法においては、核酸の相補的塩基対をモデルとして、高い分子認識能をもつ合成高分子を合成することに成功した。また、農薬を認識する高分子をモレキュラーインプリンティング法によって合成し、その分子認識能を評価して、合成した高分子が高い分子選択性を示すことを確認した。モレキュラーインプリンティング法の確立とまでは言わないまでも、めざましい成果が得られたと言えよう。
2.については蛋白質と合成高分子をハイブリッド化し、有機溶媒中でも電子伝達機能を持つ化学修飾酵素を作製することに成功し、その電気的特性の評価が進んでいる。
更に生体機能分子の設計・合成の展開として生体模倣技術を用いたアプローチ、特殊な微生物を利用したアプローチも進んでおり、かつ合成した生体機能分子の解析のための技術開発も進んでいる。
