極東におけるライム病の疫学的調査、並びに伝播機序の解析

研究概要

中国、韓国においてマダニ、野鼠の捕獲調査を実施し、これらからのボレリアの分離を行った。また、すでに得られていた極東ロシア分離株についても併せて分子生物学的性状の検討を行った。
中国東北部、極東ロシアのシュルツェマダニ由来株は、Borrelia gariniiまたはB.afzeliiと同定された。また、B.gariniiでは欧州型とは別に日本に高頻度でみられる極東型が優占種であった。一方、韓国産タネガタマダニよりB.garinii、B.afzeliiの分離に初めて成功した。すでに韓国研究者によりタネガタマダニより分離されたB.afzelii関連群ボレリアの遺伝学的性状解析を行い、欧州のlxodes ricinus由来B.valaisiana、ならびに日本のハシブトマダニ由来Am501株に近縁であることを明らかにした。中国南部揚子江流域では野鼠Apodemus spp.とRattus spp.よりボレリアの分離に成功し、Apodemus spp.の一部は韓国にみられるB.afzelii関連群ボレリアであること、またRattus spp.由来株の多くは既知の種と異なり新種である可能性を示した。また、一個体のヤマトマダニより同じく新種と思われるボレリアの分離に成功した。現在、これらの株のより詳細な遺伝学的性状解析を進め、新種として記載する予定である。
主要表層蛋白質A,B,Cの塩基配列に基づく系統解析を行い、日本と欧州間で極東ロシア、あるいは中国東北部を介してB.gariniiとB.afzeliiが伝播したことを示唆する結果を得た。
