研究概要

今回、主に下記の5種類の実験について要約する。1、Polysphondylium pallidumの細胞間接着タンパク質gp64の遺伝子破壊株の作製については、従来Dictyosteliumで報告されている形質転換効率よりも数倍効率の高い条件を見出した。この条件でも、まだ、gp64遺伝子の相同的組換え体を分離していないが、現在精力的に転換体分離の努力を続けている。なお、Dictyosteliumの細胞接着タンパク質contact site Aの相同的組換え体はマックス・プランク生化学研究所より既に入手している。2、アンチセンスRNAを用いて、Polysphondylium pallidumの接着タンパク質gp64の発現を抑制した形質転換体が分離されたが、この株の接着能を測定したところ、まだ数十パーセントに相当する細胞接着能を保持していた。そこでこの細胞株を出発材料に、細胞膜を分離し15羽のウサギに免疫して抗血清を得た。この血清はPolysphondylium細胞の接着を強く阻害することから、第2の接着タンパク質を同定する具体的な手だてを得た。現在この抗体をプロテアーゼで抗体断片(Fab)に低分子化し、第2の細胞接着分子を同定する実験を進めている。3、接着タンパク質gp64上のアスパラギン結合糖鎖の改変のための部位特異的変異誘導法の確立については、基本的な技術は確立した。しかし、まだ6個の糖鎖全てについて改変した形質転換株を分離しておらず、引き続き分離を続ける必要がある。4、Polysphondyliumの細胞接着タンパク質gp64をDictyostelium株に発現させ、細胞膜上に保持させる実験系では、発現されたgp64上の糖鎖が母株と異なること、またgp64のコンフォメイションも異なる可能性が示唆されている。今後、disulfide isomeraseの遺伝子導入等によりS-S架橋構造形成能を高める努力を進める必要がある。5、MAPカイネースの欠損変異体(erk2-)は単独では集合しないが、野生株と混合すると正常に形態形成した。その際、移動体の前部(予定柄細胞域)を占めた。このことはERKタンパク質が選別に関与することを示唆している。細胞選別には細胞接着と走化性など細胞運動が重要な役割を果たしていることを考えられ、走化性検定を試みた。その結果、野生株と同様にerk2-もcAMPに対しても走化性を示すが、その感度は野生株よりも高いこと、反対にERK2の過剰発現株では低いことが明らかとなった。
