研究概要	原田は、酵母のアクチン関連タンパク質Act3pがin vitroでコアヒストンと相互作用する能力を有し、また酵母の核内でコアヒストンとAct3pの両者を含むタンパク質複合体が存在することを示した。また、スティルマンと共同で、Act3pの温度感受性変異株において、遺伝子発現とクロマチン構造の両方に変化が現れることを示した。これらの結果は、Act3pがクロマチン構造の維持・変化を通じて遺伝子発現調節に関与する新規のアクチンファミリータンパク質であることを示している。また、Act3pに相同性のあるヒトcDNAクローンを得て、現在解析を行っている。ヴインターベルガ-は人工変異ACT3遺伝子を作製し、これを酵母に導入して表現形を解析することで、Act3pの核移行に必要な領域を同定し、また、Act3pにATP結合能があることを示した。ステイルマンは、Act3pの点変異とともに合成致死を示す遺伝子を検索しており、現在いくつかの候補を得ている。ク-パ-はTwo hybrid systemを用いてAct3pのフイラメント形成能を調べており、現在までのところ、Act3pがフイラメントを形成しないことを支持する結果をえている。本年度は、ウイーン大学ガン研究所のヴインタースベルガ-教授を招へいし、原田と共に変異ACT3遺伝子を作製した他、東京医科歯科大学難治疾患研の白形正樹博士、京都大学理学部の柳田充弘博士および大阪大学大学院工学研究科の金谷茂則博士を訪問し、Act3pの3次元高次構造の推定や、その高次構造と機能の相関についての討論を行った。また、原田を、ワシントン大学、ユタ大学、ウイーン大学に派遣し、各大学において研究分担者との共同実験ならびに研究打ち合わせを行った。
