筋形成におけるミオシン結合蛋白質の役割

研究概要	横紋筋特異的なミオシン結合蛋白質であるC-蛋白質はミオシンの集合の制御に関わる一方、アクチンにも作用して収縮機能に影響を与え、さらに弾性繊維蛋白質コネクチンに結合しサルコメアの構造形成に寄与するとされる。最近、心筋C-蛋白質の突然変異が家族性肥大形に筋症の一因となることが知られていたが、このことは筋の機能維持におけるC-蛋白質の重要性を示している。本研究では筋収縮構造の形成におけるC-蛋白質の役割を解明するために、C-蛋白質の構造及び発現を解析、さらに筋原線維形成の制御に働く他の因子についても検討し以下の成果を得た。1)マウス速筋、遅筋C-蛋白質のcDNAを分離、全構造を決定した。2)ノーザンブロット法によりマウスの筋形成過程でのC-蛋白質の発現を解析、心筋では発生過程を通じて心筋C-蛋白質のみを発現骨格筋では遅筋型から速筋型への変換が起こることを見出した(トリでは胚骨格筋に心筋C-蛋白質が現われるが、マウスでは現れない)。3)C-蛋白質アイソフォームを識別する抗体を用いて蛋白質レベルで更に解析し、上記2)の事実を蛋白質レベルでも確認した。4)心筋C-蛋白質には骨格筋C-蛋白質にはないリン酸化部位が存在するが、ニワトリ心筋C-蛋白質にはリン酸化部位を含む15アミノ酸残基の配列の有無により異なる2種のアイソフォーム(I型、II型)の存在を見出した。この15残基を特異的に認識する抗体を作成、I型とII型の発現、局在の違いの有無を検討し、心筋ではI型、II型の発現が示唆されたが、胚骨格筋ではI型がほとんど発現されないことを解明した。5)筋細胞でアクチン制御に係わる因子コフィリンの発生過程での量的な変化およびリン酸化による機能制御を明らかにした。6)ヒト肥大型心筋症で見出された変異C-蛋白質が筋細胞に及ぼす影響を探るために、遺伝子操作によりトリ心筋C-蛋白質遺伝子に同様な変異を導入することに成功した。次年度、この変異遺伝子を心筋および骨格筋に導入して変異蛋白質を発現させ細胞の表現形質の変異を探る(日米の共同プロジェクト)
