研究概要

終止コドンの認識からリボゾーム複合体の解離に至る翻訳終結の仕組みは、分子生物学の基本的な問題であるにもかかわらず、その分子機構が今日まで明かにされていない。本研究では、翻訳終結の基本的な分子機構を解明する目的で、ペプチド鎖解離因子の機能・構造、リボソームとの相互作用、転写後調節における役割等を解析した。その結果、ペプチド鎖解離因子は、原核生物から真核生物に至るまで普遍的なモチーフを保存し、その蛋白質ドメイン部分がtRNA分子の構造と酷似することを、生化学的および分子遺伝学的に明らかにした。特に、バクテリアでは2種類の解離因子(RF1,RF2)が異なる終止コドンを識別できるが、この選択的な終止コドン認識特性が変化する突然変異を分離することに成功した。これは、tRNAのアンチコドンに対応する蛋白質のアンチコドン部分が存在して終止コドンを直接認識することを裏付けるものである。この「tRNAもどき」の解離因子という考えは。何故解離因子が終止コドンを識別できるのという、分子生物学創設以来の難問を簡単明瞭に説明し、あわせて「分子擬態」(Molecular Mimicry)という新しい概念を生物学に提出するものであり、その学術的意義は極めて大きい成果といえる。今後は、構造生物学的な立場から、分子擬態研究を推進する予定である。さらに、グアニンヌクレオチドに依存して解離反応を促進する制御因子(RF3,eRF3)が原核生物、真核生物に普遍的に存在し、解離因子のリサイクリングに係わることを明らかにした。このうち、出芽酵母から分離されたeRF3は動物細胞のプリオンと同じ特性を持つプリオン様タンパク質であるという、驚くべき事実も明らかになった。その生理的な意義は、現在のところ不明であり、今後の研究によって解明したいと考えている。
