研究概要

終止コドンの解読は、分子生物学の基本的な問題であるにもかかわらず、その分子機構が今日まで明かにされていない。我々は、終止コドンの認識を触媒するベプチド鎖解離因子が原核生物から真核生物に至るまで普遍的なモチーフを保存し、その蛋白質ドメイン部分がtRNA分子の構造を擬態することを明らかにしつつある。この「tRNAもどき」の解離因子という考えは、蛋白質による終止コドン識別の仕組みを簡単明瞭に説明しうる新しい概念である。翻訳終結の分子機構の解明と分子擬態モデルを検証する目的で実施した研究成果を以下にまとめる。
1. 解離因子のtRNA擬態ドメインに保存されるアミノ酸をシステマチックに改変し、アンチコドンとして終止コドンの選択的な識別に必要不可欠な5つのアミノ酸の組みを同定した。さらに、解離因子リポソーム複合体上でのそれらの残基のトポロジーをフットプリント分析によって明らかにした。
2. 原核生物RF2の1アミノ酸置換(E167K)によって、3種類の終止コドンに対して全能的な終結能を獲得した変異体を昨年分離したが、この近傍位の他のアミノ酸置換が同様に全能性をもたらすことが明らかとなり、終止コドン3文字目の選別機構の理解が進んだ。
3. 解離因子の構造解析を目的に、高度好熱菌から解離因子eRFlを分離し、結晶化を行った。その結果、三方晶系結晶の作成に成功し、X線回折データ測定から空間群はP3_1またはP3_2、格子定数がa=b=64.9,c=86.9Åであり、非対称単位(結晶格子内の独立な領域)にRFlが1分子存在することが明らかになった。
4. 分裂酵母の解離因子eRF3・eRFl相互作用部位を詳細に明らかにした。
