ミオシン-植物細胞における役割と局在の確定

研究概要	我々はこの一年間Arabidopsisを用いて、後に交配で使用する、緑蛍光性タンパク質-小胞体(Green Fluorescent Protein-Endoplasmic Reticulum=GFP-ER)の特徴を解明した。そして、成長の初期段階に見られる新たなタイプの小胞体を観察した。この時期に見られる小胞体は、板状でいくつもの細がい孔があり、これが、少しずつ変化し、細胞成長後期には、良く知られている細胞皮質の網状の小胞体に変化する。更に、根毛についても新たな観察があり、細胞皮質の小胞体は、根と茎の表皮細胞の小胞体との中間型であった。そして、他の細胞と同様に細胞成長後期には、網状の小胞体に変化する。これらの結果は、我々の重要な功績であり、現在論文として国際的な科学雑誌に投稿中である。植物の小胞体の動きは、アクチン-ミオシンによってコントロールされている事は確かであるが、どのミオシンアイソタイプであるかを、つきとめる上で、アンチセンス植物との交配を行わなければならない。そこで、我々は、Arabidopsisのアンチセンスミオシンを単離し、GFP-ER Arabidopsisとの交配を開始した。この仕事は、現在進行中である。これらのいくつかのアンチセンス植物には、はっきりとした表現型が見られ、国際的な学会でこの事について発表を行った。更に、ArabidopsisのGFP-ミトコンドリア膜タンパク質においても、GFP-ER植物と同様の実験を始めた。ホモザイガスの系統が単離されしだい、アンチセンス植物との交配を行う考えである。
