樹状細胞の機能とその特殊性に関する研究

研究概要	強力な抗原提示細胞として免疫応答の誘導に関与することが知られている樹状細胞は、分布域によって分化成熟段階が異なり、抹消において異物を捕食・分解し、所属リンパ器官のT細胞領域へと移動して特異的T細胞を活性化することが知られている。ところが定常状態におけるリンパ器官T細胞領域に分布するいわゆる相互陥入樹状細胞(IDC)はその表現型のみならずT細胞活性化における機能も異なることが示唆されている。そこで本研究は、末梢組織のいわゆる未熟な樹状細胞の性状を解析すると同時に、マウスリンパ節におけるIDCの調製法を確立し、その機能を検討することを目的として行い、以下の結果を得た。 1.表皮内ランゲルハンス細胞や骨髄由来樹状細胞では、MHCクラスII分子は主に細胞内に存在し、この時に捕食した抗原を消化分解してMHCクラスII分子に結合した後、細胞表面へと運搬し、T細胞に提示することが、生化学的ならびに細胞生物学的検索により証明された。 2.IDCの精製法を確立し、その結果、定常状態のリンパ節にはCDllc、DEC-205等の発現強度から少なくとも2-3種類の異なる表現型をもつ樹状細胞が存在することが確認された。また、これらの細胞は内因性抗原ペプチドを多く発現しており、特異的T細胞の活性化と同時にアポトーシスを誘導することも示されたことから、末梢リンパ器官における自己免疫寛容の誘導に関与することが示唆される。 3.筋肉内あるいは皮下に抗原決定基をコードするDNAを接種すると、特異的免疫応答が誘導されるが、局所あるいは所属リンパ器官より調製される樹状細胞内にDNAが取り込まれていることが明らかになり、これらが抗原提示細胞として働いていることが示された。
