動脈硬化症を中心とした心血管系における遺伝子治療法の開発

研究概要

平成8年度は、アデノウイルスを用いた遺伝子導入法が循環器系にどのように適用可能かを、種々の初代培養細胞を対象に検討を加えた。
その第一は、レポーター遺伝子としてβガラクトシダーゼ遺伝子を組み込んだ5型アデノウイルスを用いた。初代培養細胞に血管内皮細胞、新生児ラットからの心筋細胞、アダルトラットからの心筋細胞いずれにおいても、アデノウイルス感染後、翌日より非常に良好なβガラクトシダーゼ活性を検出することが出来た。
従来のアデノウイルスベクターは、組み換え体の作成、クローニング、組み込む遺伝子のサイズに煩雑さや制限があったため、これらの欠点を改良したベクターの開発が望まれていた。これに関しても、共同研究者のDr.Gerard R.D.らとの共同研究により、ポリリジン修飾アデノウイルスの有用性を検討した。この修飾ウイルスは、外来プラスミドを細胞内に導入させることが可能であったが、ポリリジン修飾の効率及び遺伝し導入効率に変動が大きいため、さらにより簡便なベクターを開発中である。現在、これらと同等の遺伝子導入効率を得られる非修飾coniugatedアデノウイルスが、まだpreliminaryであるが開発されている。
また、循環器系においては、心筋炎、心筋症において、サイトカインの関与が重要であることが次第に判明してきた。これらのファクターをmodulateすることで心筋症、心筋炎、心機能などを改善、治療できる可能性が開けてきた。これらの基礎的、臨床的研究が進んでいる。(別掲論文集参照)
