研究概要

アポトーシスの分子機序解明を目指し、我々はこのプロセスに関わる遺伝子、ICEファミリ遺伝子やbc1-2ファミリ遺伝子などの解析を行ってきた。これら遺伝子産物の生化学的機能の解明および新たなアポトーシス関連因子の同定には、in vitro系は極めて有用な方法の一つである。
現在多くの研究室で利用されているin vitroアポトーシス系は、Yuri Lazebnikが開発したものをベースにしているが、この系ではアポトーシスの特長である核変性が顕著な形でみることが出来ないため、多くの場合アポトーシスの付随現象であるDNA切断をアッセイマーカーとして用いられている。我々は、この難点を克服する新たなin vitro系を確立した。この系ではアポトーシス特有の核変性が顕著にかつ再現性よく観察されるほか、アポトーシスに必須であることを最近我々が示した能動核輸送も必須であり、極めて生細胞に近いin vitro系といえる。この系は、既存のアポトーシス関連因子の機能解析および新たなアポトーシス関連因子の探索に極めて有効である。
一方、アポトーシス抑制遺伝子bc1-2の生化学的基盤を明らかにする目的で、単離ミトコンドリアを用いたアポトーシス解析系を確立した。我々は、細胞を用いた解析から、アポトーシスの進行にミトコンドリアの脱機能(膜電位の低下)が重要な役割を果していること、およびその脱機能をBc1-2が抑制することを明らかにしたが、単離ミトコンドリア系でこのBc1-2機能を再現することに成功した。この系はBc1-2の生化学的機能の基盤の割り出しに極めて有効である。
