細菌・動物細胞間接合による癌遺伝子の伝達系の確立

研究概要	SV40のoriとプロモーターを持ちレポーターとして青色蛍光発色蛋白質GFP遺伝子とネオマイシン耐性Neo遺伝子を選択マーカーとする動物細胞用接合伝達プラスミドpBASreenを構築し、このレポーター遺伝子がCOS1やNIH3T3細胞で効果的に発現するとともに接合伝達遺伝子も機能することも確認することができた。しかしながら、大腸菌から動物細胞への接合伝達で問題が生じた。それは、接合伝達のためにプラスミド受容細胞の動物細胞と供与細胞の大腸菌を混ぜると大腸菌の増殖速度が非常に大きく早く増え過ぎてしまうことと大腸菌が培養細胞に対してトキシックであることである。この問題は、分裂時に核を欠くミニセルを産製する大腸菌突然変異体を使い解決した。また、ミニセルを選択的に濃縮する方法を開発するとともに、ミニセル自身にも生物界間接合能があることを証明した。一方、土壌細菌アグロバクテリアはその巨大Tiプラスミド上の植物オンコジーンを植物細胞へ伝達移行させて腫瘍を形成させることが知られている。大腸菌と違いアグロバクテリアは真核細胞に対して毒性が少ないことを見出した。この点とオンコジーン伝達の共通性からTiプラスミドのゲノムの構造解析も平行して行った。最初のTiの206、482bp決定と解析の結果、Tiを接合伝達させるtra遺伝子系とオンコジーンを伝達させるvir遺伝子系とが遺伝子構造上完全に独立した構造になっていることが解った。このことは、癌遺伝子の動物細胞への接合伝達を考える上で重要な示唆を与える。
