研究概要	標的遺伝子組換えの有用性は手法が開発されて10年にも満たない現時点で、既に生命科学分野の重要課題である発生・発がん・免疫・神経系など多くの基礎的研究、医学的応用面での研究にも大きく貢献してきている。現在ではレポーター遺伝子の挿入や微細な変異の導入も可能となりさらに応用の幅が広がっているが、標的遺伝子組換えの最初の段階であるターゲティング・ベクターの構築はいまだに時間的な制約となっている。この点を解決しさらに系統的かつ簡便に点突然変異などの微細変異の導入を目的としたターゲティング・ベクターを構築するシステムを開発することが本研究の目的である。ネガティブ選択マーカーを組み込んだファージ-プラスミドのベクター系を開発した(投稿準備中)。アンバー3重変異を持つλSyrinxファージを用い、アンバー変異を抑制するSupF遺伝子断片とポジティブ選別用マーカーのneo耐性遺伝子の融合カセットを構築して系を確立した。系の開発段階初期では、これまでの研究で標的遺伝子組換えにつき最も詳細なダーターがあるhprt遺伝子座をターゲットとした。9月にRancourt博士らを招聘して検討を加え、cRABP1遺伝子についてもneo及びneo-lacZ(レポーター)遺伝子を導入することができた。この系の改良により、効率良くしかも系統的に点突然変異などの微細変異の導入を目的としたターゲティング・ベクターを構築するシステムが開発できる見通しがたった。具体的な応用としては、fosB及びΔfosB遺伝子の発現改変ベクターを作製することとし、この遺伝子の脳領域での発現に詳しいオタワ大学のRobertson博士を招聘して相談した。 2月に續がゴ-ドン研究会議に出席して開発初期段階のバ-ジョンの成果を発表した。その後、續・早川がカルガリ-大学を訪れ、新しく開発した系によるターゲティングベクター構築ラ-ブラリー作製の予備実験を行った。本研究による相互訪問の過程(共同実験・セミナー・ディスカッション)で、續のグループの大学院生の一人がRancourt博士の共同研究者のもとへ留学することに決まった。
