研究概要

標的遺伝子組換えの有用性は手法が開発されて10年にも満たない現時点で、既に生命科学分野の重要課題である発生・発がん・免疫・神経系など多くの基礎的研究、医学的応用面での研究にも大きく貢献してきている。現在ではレポーター遺伝子の挿入や微細な変異の導入も可能となりさらに応用の幅が広がっているが、標的遺伝子組換えの最初の段階であるターゲティング・ベクターの構築はいまだに時間的な制約となっている。この点を解決しさらに系統的かつ簡便に点突然変異などの微細変異の導入を目的としたターゲティング・ベクターを構築するシステムを開発することが本研究の目的である。
ネガティブ選択マーカーを組み込んだファージ-プラスミドのベクター系を開発した。アンバー3重変異を持つλSyrinxファージを用い、アンバー変異を抑制するSupF遺伝子断片とポジティブ選別用マーカーのneo耐性遺伝子の融合カセットを構築して系を確立した。系の開発段階初期でのhprt遺伝子座をターゲットとした予備実験の結果を踏まえ。cRABPI遺伝子についてもneo及びlacZ-neo(レポーター)遺伝子を導入することができた(印刷中)。この系の改良により、効率良くしかも系統的に点突然変異などの微細変異の導入を目的としたターゲティング・ベクターを構築するシステムが開発できる見通しがたった。具体的な応用としてfosB及びΔfosB遺伝子の発現改変ベクターを作製することとし、平成9年夏に績・中別府がカルガリ-大学のRancourt博士並びにこの遺伝子の脳領域での発現に詳しいオタワ大学のRobertson博士の研究室を訪れて相談した。平成10年1月にRancourt博士を招聘し、今後の具体的な戦略を立てた。3月下旬に中別府がカルガリ-大学を訪れ、新しく開発した系によるターゲティングベクター構築ラ-ブラリー作製の進展状況の把握並びに、ES細胞の操作について共同実験をすることにしている。本研究による相互訪問の過程(共同実験・セミナー・ディスカッション)で、新たにカルガリ-大学のRiabowol博士のグループとINGI遺伝子について共同研究することに決まり、平成10年度からスタートする国際学術共同研究で展開したいと考えている。
