研究概要	心臓障害に関する新規遺伝子を分離することを目標に21番染色体および22番染色体の疾患関連領域のDNAコンティグを作成し、エキソントラッピング、ゲノムシーケンシング、cDNAスクリーニングなどの手法を併用しながら解析を進めてきた。これまでに、21番染色体からSIM、MNB、KNP1、KNP3、KNP4などいくつかの新規遺伝子を分離した。特にSIM遺伝子に関しては、アントナラキス教授との共同研究によってSIM1/SIM2という2種類の類似遺伝子を発見し、それらの構造、組織発現パターンなどを解析した。これらの遺伝子は脳の発生を制御する転写調節因子として作用していると考えられた。一方、胎児心臓に特異的な発現を示すcDNAクローンも分離したが、未だ遺伝子の全容は明らかになっていない。さらに、22番染色体ではマクダ-ミット教授との共同研究として新規の遺伝子1個を同定した。この遺伝子はアポトーシスの制御に関与するといわれているマウスBid2のヒト型ホモローグで、心臓の形態形成の過程である種の細胞の出現や消失を制御している可能性もある。このように、当初の目的は順当に進んでいるが同時に全く予定外の進展がみられた。すなわち、21番染色体にマップされていた遺伝性の自己免疫疾患APECEDの原因遺伝子を発見することができAIREと名付けた。AIREは転写調節因子で、免疫寛容におけるT細胞の成熟過程などを制御していると推定した。この研究は、当教室が見出した候補遺伝子に関して、アントナラキス教授および新たにフィンランドのクローン教授の協力を得て患者血液DNAを解析し、いくつかの変異を検出して原因遺伝子と確認することができた。2月には両教授を招聘し自己免疫疾患に関するミニシンポジウムを開いて70人余の聴衆とともに公開で意見交換した。滞在中の討論で今後のさらなる共同研究の方向付けができた。
