NODマウスの抗原提示機能に関する分子遺伝学的研究

研究概要	IDDMのモデル動物であるNODマウスでは少なくとも16の関連遺伝子座(Idd)が報告されているが、それらの変異の実体や相互関係は、MHC classllに連鎖しているIdd 1以外は不明である。本研究では、抗原提示機能とT細胞の反応に関連する遺伝子と他Iddを含むNODマウスの変異遺伝子の連鎖を解明することを目指し、以下の結果を得た。(1)MSM由来のIdd3及びIdd4導入NODコンジェニックマウスを用いてinsulitisを指標にして解析したところ、Idd3はIL-2遺伝子を含む領域内にマップされた。しかも、NODのIL-2をコードする遺伝子配列はMSMと異なっていた。抗CD3抗体によるTCR複合体の刺激を受けたT細胞からIL-2産性能はNODマウスで低下していた。これらの結果、IL-2はIdd3の候補遺伝子であることが、強く示唆された(論文準備中)。(2)ゲノム中へのレトロウイルスの挿入が遺伝子の変異原となる可能性を考え、NOD中の内在性レトロウイルスについて検索した。その結果、第18染色体に挿入されたC型レトロウイルスのEnv遺伝子断片は、抗原提示に重要なIi遺伝子の1cM以内にマップすることを突き止めた。しかし、その挿入はIi遺伝子の構造や発現制御に直接影響を与えうるほど近傍に位置するという根拠は得られなかった。このほかC型レトロウイルスのEnv遺伝子断片はいずれも既知のIdd遺伝子とは異なる位置に挿入されていた。これらの中には免疫関連遺伝子の近傍に位置するものもみられ、その構造や発現に影響を与える可能性が考えられ、更なる解析が必要である。今年度の研究により、IDDM関連遺伝子座の内Idd3の候補遺伝子としてIL-2を同定した。一方、NODマウスに特異的レトロウイルスEnv遺伝子断片の18染色体への挿入はIi遺伝子の近傍ではあったが、その発現に影響するとの証拠は得られなかった。むしろ、IL-2産性能の障害によりIi発現誘導が抑制された結果と考えられ、さらなる解析を予定している。
