研究概要	i)ラット初代培養肝細胞に於けるTNF-α遺伝子と初期応答遺伝子の発現ノジュラリンによる肝発がん二段階実験の際、肝組織にTNF-α mRNAとjun及びfosファミリー初期応答遺伝子の発現が強く誘導されることを昨年報告した。TNF-αは通常マクロファージから遊離されると推測されている。私共はTNF-αを内因性発がんプロモーターと考えているから、肝細胞がTNF-α mRNAを発現し、又、同時に初期応答遺伝子の発現も誘導するか検討した。ラット初代培養肝細胞にノジュラリンを処理すると、著明なTNF-α mRNAの発現亢進とその安定性が認められ、ラットで得られたと同様の結果が認められた。初期応答遺伝子に関しても同様であった。この結果から標的細胞である肝細胞は、ノジュラリンの処理によりTNF-α mRNA、及び、初期応答遺伝子が発現することを証明した。 ii)肝発がんプロモーションに於けるTGF-βレセプターの発現変化肝発がん二段階実験で得られたGST-P陽性巣について、TGF-βレセプター、タイプ1とタイプ2の発現を検討した。前がん病変の組織は、明らかにタイプ1とタイプ2の発現が減少していた。この結果は、ノジュラリンの発がんプロモーションの経過と、TGF-βレセプターの反応性の減少が、密接に関連あることを示唆した。 iii)マウス皮膚発がんプロモーションとTIS21遺伝子発現平成7年度第1回アジュー大学-埼玉県立がんセンター研究所の合同研究会に於いて、林所長のグループがTIS21遺伝子について長年研究していることを知った。しかも、TIS21は新しいがん抑制遺伝子として作用することが推測された。私共の研究グループは長年、マウス皮膚発がんプロモーションについて研究している。この皮膚発がんプロモーションにより生じる腫瘍とTIS21遺伝子発現との変化を検討することにした。アジュー大学医学部生化学講座Myung Soog Lee助手を受け入れこの研究に従事するよう取り計らった。いろいろと処理したマウス皮膚からRNAを精製し、TIS21遺伝子の発現を定量的RT-PCRを行った。正常な皮膚ではTIS21 mRNAは発現していたが、100mgジメチルベンツアントラセン(DMBA)を処理すると、TIS21 mRNAの発現は抑制された。腫瘍としてパピロームとがん腫では、TIS21 mRNAの発現には量的差は認められなかった。興味があったのは発がんプロモーター、オカダ酸の単独処理で、TIS21 mRNAの発現が亢進することが見出された。
