研究概要

各研究分担者の平成8年度の研究成果は以下の通りである。
1)ヒラメおよびニジマスのトランスフェリンcDNAをクローン化し、構造解析を行った。ニジマスのトランスフェリン遺伝子をPCR後、制限酵素断片長多型解析を行ったところ3タイプに分けることが可能であった。
2)カラフトマスの主要組織適合抗原クラス|のcDNAをクローン化し、構造解析を行った。PCRによる多型解析を行い、カラフトマスの主要組織適合抗原の多型を検出することができた。ウナギの主要組織適合抗原クラスBのcDNAおよびメダカの主要組織適合抗原クラス||のcDNAをクローン化した。
3)耐病性遺伝子の特性などを解明するためのトランスジェニック魚を効率よく作製するための手法として精子を脱水後、エレクトロポーレーションを行う方法を開発した。
4)グラミ-のミトコンドリアDNAのDループ領域の多型性を解析したが、顕著な地域特異性は見られなかった。
5)病原微生物に感染したヒラメの白血球で特異性に発現する遺伝子が存在することをディファレンシャルディスプレイ法により確認した。
6)コイの白血球に対するモノクローナル抗体を作製し、栓球やリンパ球を識別できるようになった。
7)サケ科魚類の外部寄生虫ギロダクイルスの地域変異・遺伝的多型性が見られることをRAPD法により明らかにした。
8)魚類病原菌のみならず変温動物にも病原性の認められた運動性エロモナスの遺伝子解析を行い、新種である可能性が示唆された。
9)魚類病原細菌をEdwardsiella tardaの溶血素遺伝子をクローン化し構造解析を行ったところ、溶血素を活性化し、菌体外へ分泌する遺伝子を連座して存在することが明らかとなった。
