窒素酸化物/オゾン系の複合化学効果の精密解析と有機合成への応用

研究概要	車両の排気ガス中に比較的多量に存在する縮合多環系芳香族ケトンのベンズアントロンおよびフェナレノンについて、単独ならびにオゾン共存下における二酸化窒素との反応を種々の条件下で検討し、生成物の分布を調べるとともに、エイムス試験によりそれらの変異原性を検討した。3-ニトロベンズアントロン(3-NBA)の場合に著しく強い変異原性が認められ、ラットを用いた実験では、短期間の継続的吸入により肺気道内に腫瘍の発生が確認された。3-NBAとDNAの反応を調べた結果、一般の芳香族二トロ化合物が生体内還元過程でN-C結合によりプリン塩基の8位へ結合するのに対し、3-NBAはC-C結合で同位置へ結合することが分かった。生体内で発生したニトレニウムイオン中間体が、隣接するカルボニル酸素との電気的な相互作用を通じて安定化されるため、この特異性が発現したものと推定した。しかし、異常に高い発ガン性との関係は、現在のところ不明である。フェノール、ジメチルアニリンに代表される活性芳香族化合物と、スーパーオキシドの共存下における一酸化窒素との反応の動力学と生成物を検討した。ニトロフェノールとカテコールが主生成物として得られ、反応速度とHammett定数や酸化還元電位との間によい直線相関が認められることや、ナフトキノンがエポキシ化され、ジメチルアニリンが脱メチルされることなどから、反応はペルオキシナイトライトのN-C結合の開裂で発生したニトロソニウムイオンによる-電子酸化を経て進むものと推定した。
