研究概要

NO合成酵素およびキサンチン酸化酵素はともに分子量30万前後の2量体蛋白質であり、複数の反応中心をもつ複合金属フラビン酵素である。本研究ではこれらの複合金属酵素に加えセロビオース脱水素酵素(ヘムフラビン酵素)、およびラット由来ヘム結合蛋白質(HBP23)につきその構造と機能の解析を課題とした。研究経過とその成果は以下のとおりである。NO合成酵素 : NO合成酵素ヘム結合ドメインはシトクロムP450と分光学的に類似していることが示唆されていたが、NO生産の場であるヘム中心の微細環境については十分解明されていなかった。可視、電子スピン共鳴、X線吸収スペクトル法により、基質非結合型、アルギニン結合型、ヒドロキシアルギニン結合型、銅イオン阻害型マウスnNOSホロ酵素の酸化型ヘム微細環境の構造変化を解析した。NO合成酵素活性に影響を与える諸因子について検討を加えた。特に脂質の結合部位とその阻害機構を検討した。キサンチン酸化酵素 : すでに作成してあるキサンチン酸化酵素に引き続き、脱水素酵素型の精製法を改良し結晶化に成功した。立体構造が2.1Aまでのデータセットを基に構造が解かれた。さらに2つの非ヘム鉄の同定を部位特異的変異法によりおこなった。HBP23 : HBP23の結晶構造を箱嶋らと共同で明らかにした。またヘム結合能についても北川らと共同でラマン法いより解析した。HBP23はthioredoxin fold構造をとっており,酸化型酵素は二つのサブユニットから提供されている二つのシステイン残基(Cys-52、Cys-173)によりジスルフィド結合が形成される。これらの残基はファミリー内での保存性が高く、部位特異的変異を導入しperoxidase活性を解析した結果,peroxidase活性の活性中心であることが明らかとなった。
