研究概要

鉛リボザイムが生理機能を発現するには、鉛が必要であるが、その活性はもう1つの金属であるマグネシウムによって正及び負の調節を受けている事を明らかにした。CDとNMRを用いてこの調節の機構を原子レベルで研究した結果、低濃度のマグネシウムは、酵素一基質複合体を安定化する事により活性の上昇を引き起こすが、高濃度のマグネシウムは、活性発現に必須な鉛を複合体から追い出してしまう為に活性の下降を引き起こしてしまう事が明らかになった。両金属の結合部位も既に同定できた。
