研究概要	reverse polarity(repo)遺伝子産物はショウジョウバエのP-エレメントの挿入変異法により見い出されたpaired-likeホメオドメインを持つグリア細胞に特異的に発現する転写因子であり、repo機能欠損変異体における解析からグリア細胞の終分化に関わっていると考えられる。本研究はショウジョウバエ中枢神経系内での異所発現や酵母のone-hybrid法等の系を用いてrepo下流遺伝子産物を同定することにより、グリア細胞の分化決定機構の遺伝子ネットワークを明らかにすることを目的とする。本年度は以下4点の研究成果を挙げた。 (1)repo遺伝子産物の全長或いは欠失変異体をGALAのDNA結合ドメインを融合し、酵母内で発現させてGALAレポーターの転写活性化を検討して、repo遺伝子産物が転写活性因子でありホメオドメイン外のN末側及びC末側に少なくとも一箇所ずつ活性化に必要なドメインが存在することが示唆された。 (2)repo遺伝子産物のin vivoにおけるグリア細胞への分化促進機能を検討するために、神経芽細胞に異所発現させ、subsetのグリア細胞を染色するエンハンサー・トラップ系統において、部分的且つ間接的ではあるがマーカー遺伝子の発現の亢進を見いだした。現在責任遺伝子のクローニングを行っており、さらにこれらの遺伝子産物の発現がrepoに依存的或いは非依存的であるかについて、repoの上流であるglial cells missing(gcm)をrepo機能欠損変異下で異所発現させることにより検討する予定である。 (3)repo遺伝子産物の下流標的遺伝子を同定するために、酵母のone-hybrid法によりrepo遺伝子産物に依存して転写活性化される可能性のあるゲノムクローンを約50個分離した。 (4)repoと同様にpaired-likeホメオドメインを持つ遺伝子群を探索し、線虫のunc-4の相同分子(Dunc-4)をショウジョウバエより分離し、subsetの神経細胞において発現していることを明らかにした。現在Dunc-4が運動ニューロン或いは介在ニューロンに発現しているのか検討するために、poromoter領域を同定しtau-lacZとの融合遺伝子を作製している。またrepo遺伝子産物との構造機能的相同性を検討するために、転写抑制因子であるengrailedの転写抑制ドメインとの融合遺伝子を遺伝子導入してDunc-4の機能をdominant-interferする個体を作製を検討している。
