遺伝子欠損マウス作製による学習・本能行動に関与する分子機構の解析

研究概要	我々は哺乳類脳機能制御分子メカニズムを解析する目的で、現在までに遺伝子欠により哺乳行動、ミエリン形成過程、空間学習、情動行動、聴覚性痙攣発作の異常が明らかとなっているFynに注目し1)Fyn欠損マウスの薬理学的な脳機能解析、2)脳神経系でのFyn結合分子の解析、3)他の遺伝子欠損マウス作製による脳神経系での分子機能の解析を行った。その結果、Fyn欠損マウスではNMDA、カイニン酸による痙攣感受性の亢進が認められた。また、興味深いことにGABAA受容体関連アニタゴニスムによる痙攣感受性の違いが認められた。これらの結果はFynが神経伝達機能に関わっている可能性を高く示唆している。また、Fyn結合分子については現在まで7種類の分子の同定に成功しており、その内のTH82は新たなRNA結合分子であることが明らかとなった(J Neuroscience Res.,in press)。現在、他の分子については解析を進行中である。また、新たに記憶・学習で機能が予想されてるNMDAレセプター受容体のε2サブタイプ、GAD65について遺伝子ターゲティング法を用いた遺伝子欠損マウスを作製して、これらの分子が脳機能発現に重要な役割を持っていることを明らかにした(Cell、1996,BBRC,1996)。
