研究概要

昆虫の呼吸器である気管系は体内に広く分布した管状の上皮細胞のネットワークで、受動的な空気の拡散によりガス交換を可能にしている。気管の形成は外胚葉が陥入した袋状の前駆体が分枝し、となり同士の枝と融合してネットワークを形成する。融合する枝の先端にはtip cellと呼ばれる細胞が同定されており、枝の伸展と融合に重要な役割を果たしていると考えられている。
気管細胞は細胞間接着分子DE-カドヘリンにより密に接着した上皮構造を作っている。気管の融合に際してtip cellは擬足を伸ばして接触し、接触面にDEカドヘチンを蓄積させて接着する。気管の融合に先立ってDEカドヘリンが蓄積し、なおかつカドヘリンを欠損する変異体では、融合が起こらないこと(Uemura et al.,1996)からDEカドヘリンが気管の融合に積極的な役割を果たしていることが明らかとなった。
DEカドヘリンに依存した気管の融合は転写因子Escargot(Esg)を必要としている。Esgには二つの独立した機能があることが明らかとなった:一つはDEカドヘリン遺伝子のプロモーターを正に活性化する働きであり、もう一つはtip cellの接着後の擬足の運動を抑制する働きである。esg変異体での気管融合の異常はDE-カドヘリンの強制的な発現により部分的に救済されることよりDEカドヘリンはEsgによる転写制御の主要な標的であることが明らかとなった(Tanaka-Matakatsu et al.,1996)。
以上の研究により気管形成は形態形成運動の動的な側面を研究するのに適した系であることが明らかとなった。生体内での細胞の活動を追跡するために蛍光タンパク質gfpを利用した解析システムを開発した(Shiga et al.,1996)。
