感覚神経経路形成を制御する細胞間情報伝達分子群の同定

研究概要	我々は哺乳類脳神経回路形成の分子メカニズムを解析する目的で、現在までに遺伝子欠により哺乳行動、空間学習、情動行動の異常が明らかとなっているFynに注目して1)Fyn欠損マウスの薬理学的な脳機能解析、2)脳神経系でのFyn結合分子の解析、3)他の遺伝子欠損マウス作製による神経回路形成の分子機能の解析を行った。その結果、Fyn欠損マウスではNMDA、カイニン酸による痙攣感受性の亢進が認められた。また、興味深いことにGABAA受容体関連アンタゴニストによる痙攣感受性の違いが認められた。これらの結果はFynが神経伝達回路形成に関わっている可能性を高く示唆している。また、Fyn結合分子については現在まで7種類の分子の同定に成功しており、その内のTH82は新たなRNA結合分子であることが明らかとなった(J Neuroscience Res.,in press)。現在、他の分子については解析を進行中であるが、特に脳神経特異的に発現する新たなカドヘリンファミリーの同定に成功し解析している。また、新たに学習、神経回路形成で機能が予想されているNMDAレセプター受容体のε2サブタイプの遺伝子欠損マウスを作製した結果、脳神経回路形成での異常が認められた(Cell、1996)。また、この受容体がFynにより直接リン酸化されていることを現在までに明らかにしている。
