網膜神経節細胞の軸索切断によるサブタイプ選択的死の分子機構の解明-遺伝子治療を目指した基礎研究-

研究概要

哺乳類の網膜神経節細胞は、軸索切断すると細胞体も逆行性変性する。しかし、生存する細胞も僅かであるが存在する。ネコやラットを用いた我々の研究により、細胞は神経節細胞のすべてのサブタイプで均等に生き残るのではなく、一部のサブタイプに偏っていることが明らかになっている。私達はこの事実に着目し、神経節細胞のサブタイプ間で発現パターンに違いがある分子群について、特に変性や再生に深く関わっていると想定されている神経栄養因子とその受容体ファミリーに着目して研究を進めている。
BDNFの受容体であるfull length TrkB、truncated TrkB、p75NTRについて、それらの特異的なcDNA断片をラット脳からRT-PCRで単離した。それぞれのプローブが他の分子に交差反応しないことをNorthern Blottingで確認した後、それらをtemplateとしてDigoxigenin-labeled cRNA probeを作成した。ラットとネコの網膜切片についてin situ hybridizationを行ったところ、これら3つのプローブについて神経節細胞層と内顆粒層に陽性のシグナルが同定できた。神経節細胞のシグナルについて詳細に観察すると、p75NTRについては、明らかに神経節細胞と考えられる細胞のうち、陽性シグナルを示す細胞と示さない細胞が見いだされた。一方、truncatedとfull length TrkBについては神経節細胞層にdiffuseにシグナルが見いだされ、我々が得た標本の中ではシグナル陰性の神経節細胞は観察されなかった。以上の結果より、ラット、ネコの神経節細胞については、p75NTRとTrkBの両方を発現する細胞と、TrkBのみしか発現しない細胞とに分かれていることが強く示唆された。このことは神経栄養因子に対する反応性の違いを説明するものと推測された。これら神経栄養因子受容体の発現パターンの違いと、眼球内にBDNFを注入した時の細胞による効果の違い、及び逆行性変性から免れて生き残るサブタイプ、軸索再生を起こすサブタイプとの関連について今後重点時に研究を進めていく予定である。
