運動ニューロンに特異的に発現されるレセプター型チロシンキナーゼの機能解析

研究概要

運動ニューロンの生存を促進する栄養因子を探索する一つの方法として、運動ニューロンに特異的に発現する受容体を探索し、次にそのリガンドとしての栄養因子候補を見出すことを試みてきた。特異抗体を用いたパンニング法によりトリ胚の運動ニューロンを精製し、RT-PCRにより受容体型チロシンキナーゼをクローニングし、in situハイプリダイゼーションによるスクリーニングからRetプロトオナコジーンとEphサブファミリーのCek8(マウスではSek)が特異的に発現していること、Retの発現は座骨神経切断により低親和性NGF受容体と同様に変化することから栄養因子受容体である可能性が高いことを見出した。
平成8年度には、ビアコアを用いたRetのリガンド検索のため、Retの細胞外領域とヒトIgG Fc領域とのキメラ蛋白やHis-tagを付加した蛋白を作成した。しかし、年度途中にRetはGDNFとGDNF結合蛋白とに会合してGDNFのシグナルを伝えることが報告されたため、我々の作業仮説は正しいことが明らかになったもののRetの研究は中断した。
Cek8は四肢筋を支配する運動ニューロンに発現することから、他のEphサブファミリーメンバーをスクリーニングしたところCek4が体幹筋とごく一部の四肢筋を支配する運動ニューロンに発現していた。これらのリガンドとしてELF-1とRAGSが報告されたためトリcDNAをクローニングした。リガンドによる受容体の自己リン酸化はリガンドが細胞膜上に発現されるか、リガンド-Fcキメラ蛋白が抗Fc抗体によって凝集した形をとることが必要であった。このように受容体を介してシグナルが伝わる場合、運動ニューロンの神経突起伸展が阻害され、軸索成長を制御するガイダンス因子と考えられた。Cek4、Cek8は運動ニューロンの自然細胞死の磁気に対応して発現されるが、生存維持効果は観察されなかった。さらに、生後2日目のヒヨコの座骨神経を切断し、これらの分子が再発現することを期待したが、現在のところ抗体染色では再発現が見出せていない。
