グルコユルチュイドとエストロゲンの神経細胞死と生存に対する影響

研究概要	われわれの以前の研究から、ラット辺縁系の初代培養細胞でエストロゲンはカイニン酸による神経障害作用に拮抗し、なんらかの神経保護作用を持っていることが示されている。実際、エストロゲンがアルツハイマー病に効果があるという臨床報告もあるので、エストロゲンの神経保護作用についての研究は臨床的にも意義のあることと考えられる。今回の実験では、このエストロゲンの神経保護作用が、神経栄養因子の一つであるInsulin-like growth factor I(IGF-I)を介しているのではないかという仮説のもとに、実験を行った。ラットにestradiol 500μg/kgを皮下投与したところ、5-7日目に大脳皮質や海馬でIGF-I mRNAの増加が認められた。一方、IGF-IIやhepatocyte growth factorのmRNAには変化がなかった。カイニン酸12mg/kgを腹腔内投与して、神経細胞に障害を与えたラットを作成した。このラット脳内の大脳皮質や海馬にエストロゲン受容体やそのmRNAが発現していることを、それぞれ組織免疫化学やin situ hybridizationで確認した。しかし、このときIGF-I mRNAには変化がなかった。以上の結果から、エストロゲンはIGF-Iを介して神経保護作用をもたらしていることが示唆された。エストロゲンは脳の個体発生途中に神経栄養因子として働くことが知られており、神経細胞障害時にもこれと同様なことが生じている可能性が考えられた。
