細胞質分裂における低分子量G蛋白質Rhoの機能と作用機構

研究概要	Rho低分子量G蛋白質が、アクチン細胞骨格系を介して、細胞質分裂をはじめ、種々の細胞機能を制御していることが明らかにされている。私共は、出芽酵母においても、RhoのホモログであるRHO1が、細胞質分裂部位に局在して細胞質分裂を制御していることを明らかにしている。Rhoの細胞質分裂における役割を明らかにするためにはRhoの標的蛋白質を単離することが不可欠である。本研究では、出芽酵母RHO1の標的蛋白質の単離を試み、以下の成果を得た。遺伝学的方法やTwo-hybrid法を用いて、RHO1の標的蛋白質が、プロテインキナーゼCのホモログであるPKC1とグルカン合成酵素、BNI1/BNR1であることを明らかにした。さらに、私は、RHO1が、RKC1を介して遺伝子の発現を、グルカン合成酵素を介して細胞壁の合成を、BNI1/BNR1を介してアクチン細胞骨格の再編成を制御することにより、酵母の出芽過程を統合的に制御していることを明らかにした。特に、BNI1/BNR1は、マウスやショウジョウバエ等で細胞質分裂に関与していることが明らかにされている蛋白質と相同な領域(FH1、FH2ドメイン)を有しており、Rhoが細胞質分裂を制御する際に重要な役割を果たす標的蛋白質であると考えられる。さらに、私は、このBNI1/BNR1が、アクチン結合蛋白質であるプリフィリンに直接結合してアクチン細胞骨格の再編成を制御していることを明らかにした。以上の成果を得たことにより、本研究の目的は十分に達成できた。
